Alloxan is the acyl urea of pyruvate, which can be synthesized from the oxidation of uric acid. It has a special destructive effect on the β cells of the islet, terminates the secretion of insulin, and causes experimental alloxan diabetes in animals. It is used in biochemical studies.

Alloxan selectively destroys islet B cells by producing superoxide free radicals, causing intracellular DNA damage and activating the activity of poly (ADP-ribose) synthetase, resulting in the decrease of coenzyme Ⅰ content. The combined action of the two factors leads to the impairment of mRNA function and cell death. Results in the decrease of insulin concentration in the blood and the production of high blood sugar, forming insulin-dependent diabetes.
alloxan-induced hyperglycemia animal model: The success of
model preparation is affected by many factors, such as animal selection, drug dosage, route and so on. In general, rats are sensitive to alloxan, dogs are not sensitive, and guinea pigs are resistant. Modeling dose 50-70mg/kg, intravenous injection; Rat 40mg/kg, intravenous injection, administration should be rapid. At the same dose, the success rate of intraperitoneal injection is low, while the subcutaneous injection needs to be increased by 4-5 times. Fasting is another factor affecting modeling results. If rats are fasted for 48 hours, 20mg/kg intravenous injection can make most models successful. In addition, it should be noted that the aqueous solution of alloxan is unstable, and the room temperature is stable at pH3.0. It should be kept below 4 degrees at pH7.0. Some rats and mice could spontaneously relieve to normal in about 30 days. Within a few hours of modeling, some animals, especially rabbits, have hypoglycemic reactions and even die, and intravenous glucose can be used as emergency treatment. Alloxan should be freshly prepared during modeling. The difference of the average blood glucose value of each group should not be greater than 20mg/dl, and the animals whose blood glucose value does not meet the requirements should be excluded.
Studies have shown that divided small doses of alloxan have higher success rates and lower mortality than a single large dose. Therefore, alloxan is generally used in the preparation of rat diabetes model by injecting small doses in divided doses.
