| Detection Principle | Solarbio ELISA试剂盒采用基于竞争法的酶联免疫吸附检测技术。将小鼠E2抗原包被于酶标板上；实验时，分别加入梯度稀释的标准品和预稀释的样本和HRP标记的抗小鼠E2单抗，样本或标准品中的小鼠E2与包被的小鼠E2竞争HRP标记的抗小鼠E2单抗上的结合位点；形成包被抗原E2-HRP标记的抗E2单抗复合物；洗板后加入显色剂底物TMB，若反应孔中样本存在不同浓度的小鼠E2，则HRP会使无色TMB变成不同深浅（负相关）的蓝色物质，加入终止液后反应孔会变成黄色；最后，在λmax=450 nm（OD=450 nm）处测定反应孔样品吸光度（OD），小鼠E2浓度与OD450值之间呈反比，通过绘制标准曲线计算出样品中小鼠E2的浓度。 |
| Background | Estradiol is a female sex hormone produced by the ovaries，adrenal gland and also the placenta during pregnancy. Estradiol is the most important hormone during a female’s reproductive years，and is required for reproductive and sexual function as well as having an impact on the health of other organs and tissues.In females，estradiol acts primarily as a growth hormone for the reproductive organs including the vagina，the fallopian tubes，the endometrium and the cervical glands . Estradiol also enhances growth of the womb’s muscle layer，the myometrium. In addition，the hormone maintains oocytes and triggers a series of events that lead to ovulation. Though estradiol levels in men are much lower compared to those in women，estradiol has important roles in men as well. Aside from humans and other mammals，estradiol is also found in most vertebrates and crustaceans，insects，fish，and other animal species. Estradiol is produced especially within the follicles of the ovaries，but also in other tissues including the testicles，the adrenal glands，fat，liver，the breasts，and the brain. Estradiol is produced in the body from cholesterol through a series of reactions and intermediates. |
