Product Description:
Matrigel (phenol-free red) is a basement membrane matrix isolated from EHS mouse tumors rich in extracellular matrix proteins. The main components of matrigel are laminin, type IV collagen, nestin, heparin sulfate glycoprotein, growth factors and matrix metalloproteinases. At room temperature, the Matrigel (phenol red free) substrate polymerized to form a biologically active three-dimensional matrix, simulating the structure, composition, physical properties and functions of the basement membrane of cells in vivo, which is conducive to the culture and differentiation of cells in vitro, as well as the study of cell morphology, biochemical function, migration, infection and gene expression.
Cells can be grown on the surface of Matrigel (phenol red free) matrix layer with a thickness of 0.5mm, or in the three-dimensional Matrigel (phenol red free) matrix with a thickness of 1mm. Overdiluted Matrigel (phenol free red) forms a non-glial protein layer that can be used for cell adhesion, but not for cell differentiation studies.
This product is prepared from DMEM medium and contains 10ug/ml gentamicin. Please refer to the product label for specific concentrations.
Thin glue forming method:
1. After freezing and thawing, the Matrigel (phenol free red) matrix was mixed with the pre-cooled pipette tip to form a homogenate.
2. Place the required culture plate on ice and add Matrigel (phenolic red free) substrate with a concentration of 50μL/cm2 growth area.
3. Place at 37℃ for 30 minutes, ready to use.
Thick glue forming method:
1. After freezing and thawing, the Matrigel (phenol free red) matrix was mixed with the pre-cooled pipette tip to form a homogenate.
2. Place the culture plate to be used in an ice bath, mix the cultured cells with Matrigel (phenol free red) substrate, and suspend them in the substrate with the tip of a pipette gun. Add Matrigel (phenolic red free) substrate with a growth area of 200-300μL/cm2.
3. Place at 37℃ for 30 minutes, can become glue. The matrix of cell culture can be added, or the cells can be grown directly on the surface of the glue.
Thin layer coating method:
1. After freezing and thawing, the Matrigel (phenol free red) matrix was mixed with the pre-cooled pipette tip to form a homogenate.
2. The Matrigel (phenol red free) matrix was diluted with serum-free medium according to the needs of use. The optimum coating concentration was determined according to the experimental requirements.
3. The diluted Matrigel (phenol free red) substrate is coated in the desired culture vessel, covering at least the entire growing surface of the vessel. Incubate at room temperature for 1 hour.
4. Remove unbound Matrigel (phenol red free) and rinse gently with serum-free medium.
Note:
1. Since Matrigel can be gelatinized at more than 10℃, keep Matrigel (no phenol red) on ice all the time during the operation. All cell culture vessels, pipetting heads and dispensing tubes that come into contact with Matrigel (no phenol red) must be pre-cooled before use.
2.Matrigel (no phenol red) substrate will have color difference changes (light yellow to deep red), which is caused by the interaction of phenol red and bicarbonate with CO2, and the color difference will be reduced after equilibrium with 5% CO2. After freezing and thawing, gently shake the reagent bottle to disperse Matrigel (phenol free red) evenly. All operations must be carried out in a sterile environment, the reagent bottle cap can be wiped with 70% ethanol, and natural drying. A pre-cooled pipette should be used to ensure that Matrigel (phenol free red) is homogenized.
3. When dissolving, the Matrigel (phenol free red) matrix can be dissolved in ice in a refrigerator at 4℃ overnight. Matrigel (phenol free red) can be returned to liquid form after 24 to 48 hours at 4 ° C.
Operation instructions: (for reference only)
In order to ensure the gelatinization performance and stability of Matrigel (no phenol red) matrix, the dilution concentration should not be lower than 3mg/ml, which can be diluted in pre-cooled serum-free medium, and Matrigel (no phenol red) is used immediately after gelatinization.
