In Vitro | CYT997(1 μM)处理A549细胞24小时,诱导微管快速重组,包括现有的微管网络遭破坏,及一些细胞质微管蛋白在菌斑处累积,导致细胞形态发生显著变化,包括粘附细胞丢失,细胞减少。CYT997作用于16种癌细胞具有毒性,IC50为作用于HepG2细胞的9 nM到作用于 KHOS/NP细胞的101 nM。CYT997有效作用于HCT15细胞,具有多耐药机制Pgp(MDR+),IC50为52 nM。CYT997通过抑制微管聚合,使细胞周期停在G2-M分界处,且诱导磷酸化的Bcl-2增多,也提高cyclin B1表达,caspase-3激活,以及PARP的产生。CYT997处理1小时后,引起HUVEC 单层膜通透性快速且可逆的提高,IC50为~80 nM。[1] 与CYT997破坏细胞微管蛋白相一致,CYT997有效抑制增殖,诱导细胞周期停滞,且诱导人骨髓细胞系(HMCLs)和原代MM细胞凋亡。[2] |
In Vivo | CYT997口服处理大鼠的半衰期(2.5 小时)比静脉注射半衰期(1.5小时)稍长,绝对口服生物有效性为50% 到70%。CYT997口服处理给药携带 PC3移植瘤的小鼠,与Paclitaxel相比,更有效抑制肿瘤生长,这种作用存在剂量依赖性。CYT997也有效作用于携带小鼠乳腺癌4T1 细胞的同位模型,这种模型有些是抗Paclitaxel治疗的。CYT997按7.5 mg/kg剂量腹腔注射给药肝转移,在第6小时显著降低血流,与CA4P按100 mg/kg剂量处理的阳性对照作用程度相似。[1]与在体外抗骨髓瘤活性一致,CYT997每天按15 mg/kg剂量处理激进型系统性骨髓性白血病小鼠模型,显著延长寿命。[2] |
