In Vitro | IWR-1和XAV939均作为可逆的Wnt途径抑制剂并且在体外表现出相似的药理学作用。 IWR-1通过与Axin的相互作用发挥其作用,而XAV939直接结合TNKS [1]。 IWR-1(10μM)诱导β-连环蛋白破坏复合物的稳定化。将IWR-1(10μM)与MIF一起加入培养基中,细胞集落的大小极度减少,这表明在任何MIF浓度组中IWR-1都抑制MIF对NSPC增殖的促进作用。 2,5和10μM的IWR-1剂量依赖性地显著抑制NSPC的增殖。 IWR-1抑制MIF对NSPC向神经元谱系分化的促进作用[2]。在最大刺激剂量的FSH(0.5 ng/mL)存在下进行IWR-1治疗可导致FSH刺激作用的剂量依赖性抑制,在IWR-1(1μM)的最大抑制剂量下观察到> 75%的抑制作用。 IWR-1治疗部分逆转了FSH诱导的颗粒细胞CARTPT mRNA表达的抑制作用[3]。 |
