Target Point | 5-lipoxygenase;5-LOX |
Passage | Metabolic Enzyme&Protease |
Background | It is a tricyclic diterpenoid oxygen-containing compound, which can inhibit lipoxygenase activity. |
Biological Activity | Abietic acid (Sylvic acid,Abietate,Rosin Acid),an abietane diterpenoid,inhibited soybean 5-lipoxygenase with an IC50 of 29.5 ± 1.29 μM.[1-2] |
In Vitro | Abietic acid has anti-inflammatory effects by inhibiting expression of genes involved in inflammation such as those for tumor necrosis factor-a and cyclooxygenase 2 in lipopolysaccharide-induced macrophages. Abietic acid regulates PPARγ target gene expression in RAW 264.7 macrophages and 3T3L adipocytes. It can be used for regulating lipid metabolism and atherosclelosis[2]. Abietic acid is a PPARγ ligand and that its anti-in£ammatory effect is partly due to the activation of PPARγ in stimulated macrophages. In lipopolysaccharide(LPS)-stimulated macrophages, abietic acid suppresses production of prostaglandin E2(PGE2)in vitro and in vivo[1]. |
In Vivo | Abietic acid has an antiobesity effect in mice mediated by the regulation of adipogenesis[2]. |
Cell Experiment | Animal Models: Male C57BL/6J mice; Dosages: 20 mg/kg and 40 mg/kg; Administration: oral administration[2] |
Animal Experiment | pM-hPPARγ,p4xUASg-tk-luc,pCMX-CBP and pRL-CMV are transfected into CV1 cells and then,24 h after the transfection, the cells are treated with abietic acid at various concentrations for 24 h.[3] |
