| In Vitro | γ-氨基丁酸(GABA)在成熟的中枢神经系统中主要是作为抑制性神经递质。在细胞培养基中加入GABA能够促进表达GABRP的PDAC细胞的增殖，但在GABRP阴性的细胞中没有作用。而GABAA受体的拮抗剂可以抑制GABA的促生长效应。GABA的处理可促进持续性表达內源GABRP的HEK293细胞的生长。在GABRP阳性的细胞中，GABA增加胞内Ca2+的水平、激活MPAK/Erk信号[1]。GABA通过胰岛β细胞和免疫系统发挥其抗糖尿病的作用。不同于在人脑和胰岛α细胞中GABA起超极化作用，在胰岛β细胞中，GABA使细胞膜去极化，引起Ca2+内流，从而激活PI3K/Akt依赖性的生长生存信号通路[2]。 |
| In Vivo | GABA是存在于成年大脑中的主要抑制性神经递质，在免疫系统中也具有抑制作用。GABA能药物被用作治疗焦虑、戒酒、癫痫，也被用来镇静和麻醉。在脑卒中动物模型中，GABA具有神经保护作用。在外周巨噬细胞中，GABA的处理能够减少炎性细胞因子的产生。在非肥胖糖尿病小鼠模型中，GABA能减少T细胞的自身免疫和炎症反应的发展[3]。GABA由胰岛β细胞产生，它从胰岛β细胞中释放出来并作用于α细胞，使α细胞的细胞膜超极化，从而抑制胰高血糖素的分泌。STZ注射后的小鼠，在GABA处理后的53天中，具有高循环胰岛素、低的胰高血糖素、几近正常的血糖水平，代谢水平得以改善、持续稳定正常的葡萄糖水平[2]。 |
