Product Introduction:
Alkaline phosphatase (ALP or AKP) is widely distributed in mammalian tissues. The optimum pH for its activity is 9.2-9.8. This enzyme mainly exists in active substance exchange sites (cell membranes):Such as the brush-like margin of intestinal epithelium and proximal convoluted tubule of kidney, the stationary cilia of epididymis epithelium, the capillary cholangium of liver, and the endothelium of arterioles and capillary arteries. It is also found in the endoplasmic reticulum, Golgi complex, swallowing vesicles, lysosomes of intestinal epithelium, neutral granules of neutrophils and smooth muscle cell membranes.
The Alkaline Phophatase Stain Kit (Kaplow's/Azo Coupling Method) uses the azo coupling method to display alkaline phosphatase activity. The principle is that under alkaline conditions of pH 9.2-9.8, alkaline phosphatase in cells can hydrolyze substrates, release naphthol in situ, and generate insoluble colored products with chromogenic azo dyes, which are located in the cytoplasm. This kit can be used for alkaline phosphatase staining of blood, bone marrow or cell smears, frozen sections, etc. The active site is blue and located in the cytoplasm, and the results are more accurate than the metal salt precipitation method.
