Product Introduction:
Calcein, AM is a cell staining reagent that can fluorescent label living cells. After it penetrates the cell membrane and enters the cell, it is cleaved by intracellular esterase to form Calcein, which is retained in the cell and emits strong green fluorescence. Compared with other similar reagents such as BCECF, AM and CFDA, Calcein, AM showed very low cytotoxicity. The excitation and emission wavelengths of Calcein are 490 nm and 515 nm, respectively.
Calcein, AM stained only viable cells. As a nuclear dye, PI cannot cross the cell membrane of living cells. It passes through the disordered region of the dead cell membrane and reaches the nucleus, where it intercalates into the DNA double helix of the cell to produce red fluorescence (excitation: 535 nm, emission: 617 nm), so PI only stains dead cells. Since both Calcein and PI-DNA can be excited by 490 nm, both live and dead cells can be visualized by fluorescence microscopy. Only dead cells are observed with excitation at 545 nm.Calcein, AM and PI are often combined to double stain live and dead cells which are based on above characteristics.
Since the optimal staining conditions differ between cell lines, we recommend individually determining the appropriate concentrations of Calcein, AM,and PI.
