Product introduction
Alkaline protease is a proteolytic enzyme derived from Bacillus licheniformis 2709, which is bred by bacterial protoplast mutagenesis, and obtained by deep fermentation, extraction and refining. Its main enzyme component is Bacillus licheniformis protease, which is a serine type endoprotease, which can hydrolyze protein molecular peptide chain to produce polypeptide or amino acid, and has a strong ability to decompose protein. Alkaline protease is an alkaline protein with a catalytic site in serine, which has excellent properties at high temperatures and moderate alkalinity. The production process of alkaline protease adopts advanced technology such as microfiltration, ultrafiltration membrane separation, spray drying and vacuum freeze drying. The product complies with the food grade enzyme specification standards of the United States Chemical and Pharmacopeia (FCC) and the Light Industry Division Industry Executive Standard (QB/T1803-93).
Scope of application
It is suitable for hydrolyzing animal and plant protein in alkaline medium, widely used in food, medical treatment, silk degumming, leather hair removal, enzymatic detergent and other industries. The product quality has reached food grade and pharmaceutical grade quality standards.
Usage method
For enzymatic hydrolysis, the substrate concentration is 10-25%, the temperature is 50-60 ℃, the ph value is 8.5-10.5, the reaction time is 3-6 hours (can be longer or shorter according to the requirements), and the enzyme is added 0.1-0.2% (calculated by the weight of the hydrolytic solution). For other industrial and medical fields, refer to relevant industry standards.
Unit definition
Ultraviolet spectrophotometry: under the determination conditions (40℃, ph10.5), the absorbance of the TCA soluble substance released from hydrolyzed casein per minute at 275nm wavelength is equivalent to the absorbance of 1 microgram tyrosine, and the required enzyme amount is a unit of activity, expressed by u/g
Matters needing attention
Alkaline protease is a pure natural enzyme preparation, is a protein, the consumption of enzyme preparation of food as the consumption of protein-containing food is generally beneficial to the human body. For some sensitive people, such as direct ingestion of high concentration of enzyme powder or aerosol drops, it may cause allergy, and prolonged exposure may irritate the skin, eyes and mucous tissue. It is recommended to wear a mask, eye mask and other protective equipment during operation. The remaining or spilled enzyme powder should be treated in time. For a large amount of spilled enzyme powder, it should be gently swept back to the container, and a small amount should be vacuums or soaked with water to clean up. Alkaline protease is a kind of bioactive substance, which is easily affected by heavy metals and oxidants, and should be avoided in the storage process. It is recommended to store in a cool and dry environment away from light. If the storage is too long or the storage conditions are unfavorable, the enzyme activity will be reduced to different degrees. If the temperature and humidity are too high, it is necessary to increase the amount appropriately when using.
