Total bile acid (TBA) is the product of cholesterol catabolism in the liver and is the largest organic acid secreted by the liver into bile. When liver cell injury or biliary obstruction, it will cause bile acid metabolism disorder and increase serum bile acid. Therefore, the determination of bile acid can be used as an important information to judge whether the hepatobiliary system is normal or not.
Bile acids are catalyzed by 3α-hydroxysteroid dehydrogenase ( 3α-HSD ) to generate 3-oxocholic acid and NADH, which react with nitroblue tetrazolium iodide ( INT ) to form a pink substance with a maximum absorption peak at 500 nm. Based on this, the content of total bile acids can be calculated.
