Note | (1) The pre-denaturation condition of PCR procedure was 95℃ for 5 min to fully lysate blood cells and denaturate nucleic acid templates. (2) It is recommended to use high-quality primers. (3) Increase the number of PCR cycles. (4) Reduce the annealing temperature, and try the annealing temperature gradient if necessary. Extending the annealing time to 120 sec/kb (based on the maximum fragment) increases the amount of amplified product. (5) The amplification performance and specificity of primers were examined. (6) Adjust the amount of whole blood template within the range of 4%~20% final concentration. (7) The product is too long, the primer needs to be redesigned, and the product length should not exceed 1 kb. (8) Extend the extension time within the cycle. (9) Improve the use of low-yield or missing amplicon primers. (10) All primers are similar. When multiple primers contain high GC fragments, try to design primer Tm at 65~72℃ and adjust the annealing temperature at 60~68℃. (11) This product is only suitable for direct expansion, generally not for pure template DNA amplification. |

Blood Direct Multi PCR Premix has strong tolerance to blood-derived inhibitors and is a special reagent for direct DNA amplification without nucleic acid extraction of blood samples. This product has a strong inhibitor tolerance. It can directly amplify anticoagulant whole blood samples such as EDTA, heparin and sodium citrate in the presence of blood-derived inhibitors without the extraction and purification of blood sample DNA. Due to its special optimization, this product has a higher specificity and is ideal for multiple direct amplification of DNA without extraction in whole blood.
