Note | 1. The reaction of in vitro transcription is highly sensitive to RNase, please not mix Rnase in the reaction system. Experimental equipment such as pipettes and EP tubes must be RNase-free. 2. Add samples with the order in the instruction strictly. 10×Transcription Buffer should be used after returning to room temperature. DNA is easy to co-precipitate with spermidine at low temperature, which affects the transcription yield. In the preparation of the system, DNA template should be the last component added. 3. For small DNA fragments (≤300bp) in vitro transcription, it is recommended to extend the vitro transcription time to 3 hours to ensure adequate yield. It will not affect the quality of the product if extend the vitro transcription time (in 16 hours). |

T7 RNA polymerase is a DNA-dependent RNA polymerase derived from bacteriophage T7 with highly specific 5'→3' end activity of RNA polymerase.T7 RNA polymerase has high specificity for the T7 promoter and is able to synthesize large amounts of RNA using the downstream sequence of the T7 promoter as a template.
