Detection Principle | Solarbio ELISA试剂盒采用基于双抗体夹心法的酶联免疫吸附检测技术。将抗人VSIG4单克隆抗体包被在酶标板上;分别加入梯度稀释的标准品和预稀释的样本,标准品和样本中的人VSIG4会与酶标板上的包被抗体充分结合;洗板后加入HRP酶标记的抗人VSIG4抗体,该抗体会与板子上包被抗体捕获的标准品和样本中的人VSIG4发生特异性结合;洗板后加入显色剂底物TMB,若反应孔中样品存在不同浓度的人VSIG4,则HRP会使无色TMB变成不同深浅(正相关)的蓝色物质,加入终止液后反应孔会变成黄色;最后,在λmax=450 nm(OD=450 nm)处测定反应孔样品吸光度(OD),样本中的人VSIG4浓度与OD成正比,通过绘制标准曲线和四参数拟合软件便可计算出样本中人VSIG4的浓度。 |
Background | VSIG4 (V-set and immunoglobulin domain containing 4),also known as complement receptor of the immunoglobulin superfamily (CRIg) and Z39Ig,is a type I transmembrane glycoprotein. It is a B7 family-related protein and an Ig superfamily member. In contrast to the B7 family members which contain two IgG domains,VSIG4 contains one complete V-type I g domain and a truncated C-type I g domain. VSIG4 is exclusively expressed on tissue resident macrophages and binds to multimers of C3b and iC3b that are covalently attached to particle surfaces. No VSIG4 expression appears to be present in T and B cells. VSIG4 functions as a negative regulator of T cell activation,and may be involved in the maintenance of peripheral T cell tolerance,and is also identified as a potent suppressor of established inflammation. Mouse VSIG4 is synthesized as a 28 amino acid precursor that contains a signal sequence,a V-type I g domain (aa 36-115),one potential N-linked glycosylation site,and a single transmembrane domain. The V-type I g domain of mouse VSIG4 shares 86% and 8% aa sequence identity with the V-type I g domains of rat and human VSIG4,respectively. |
