| Detection Principle | Solarbio ELISA试剂盒采用基于双抗体夹心法的酶联免疫吸附检测技术。将抗大鼠IL-1R1/CD121a单克隆抗体包被在酶标板上；分别加入梯度稀释的标准品和预稀释的样本，标准品和样本中的大鼠IL-1R1/CD121a会与酶标板上的包被抗体充分结合；洗板后加入HRP酶标记的抗大鼠IL-1R1/CD121a抗体，该抗体会与板子上包被抗体捕获的标准品和样本中的大鼠IL-1R1/CD121a发生特异性结合；洗板后加入显色剂底物TMB，若反应孔中样品存在不同浓度的大鼠IL-1R1/CD121a，则HRP会使无色TMB变成不同深浅（正相关）的蓝色物质，加入终止液后反应孔会变成黄色；最后，在λmax=450 nm（OD=450 nm）处测定反应孔样品吸光度（OD），样本中的大鼠IL-1R1/CD121a浓度与OD成正比，通过绘制标准曲线和四参数拟合软件便可计算出样本中大鼠IL-1R1/CD121a的浓度。 |
| Background | Interleukin 1 receptor，type I (IL-1R1) also known as CD121a (Cluster of Differentiation 121a)，is an interleukin receptor. IL-1R1/CD121a is a cytokine receptor that belongs to the interleukin 1 receptor family. This protein is a receptor for interleukin alpha (IL1A)，interleukin beta (IL1B)，and interleukin 1 receptor，type I (IL1R1/IL1RA). IL-1R1/CD121a is an important mediator involved in many cytokine induced immune and inflammatory responses. This protein has been characterized by pharmacological and molecular techniques in the mouse brain. The spindle-shaped astrocytes enclose the wound，separating the healthy from damaged neural tissue. The shape change and subsequent repair processes are IL-1 beta activity-dependent，acting through the IL-1 type 1 receptor (IL-1R1)，as co-application of the IL-1type 1 receptor antagonist protein (IL-1ra) blocks IL-1 beta induced effects. In the spleen，a slight increase in IL-1R AcP and IL-1R1 was observed during the first hours following LPS stimulation. In conclusion，IL-1R AcP mRNA is expressed in the brain and in other tissues where IL-1R1/CD121a transcripts are found. However，the regulation of its expression is distinct from IL-1R1/CD121a. The high level of expression and the lack of regulation of IL-1R AcP transcripts in the brain under inflammatory conditions suggest that the protein might be constitutively expressed in excess. |
