Product Description | Agrobacterium EHA105(pSoup) strain was modified from EHA105 strain and was a C58 background, and its nuclear gene contained rif, a screening gene for favorable fampicin resistance. This strain carries a succinine-type Ti plasmid pEHA105 (pTiBo542DT-DNA) without its own transport function, which contains the vir gene (the vir gene is necessary for T-DNA insertion into the plant genome). The T-DNA transfer function of the pEHA105(pTiBo542DT-DNA) plasmid itself is damaged, but it can help the transferred binary vector T-DNA transfer smoothly. Some expression plasmids, such as pGreen, pGreenII-62SK and pGs2, which were missing replication-related elements (such as REP region of pVS1 replication origin or STA region of pVS1 plasmid), could not reproduce in EHA105 strain. pSoup helper plasmids can help these incomplete binary expression plasmids replicate in Agrobacterium. EHA105(pSoup) strain was tetracycline (Tet) resistant. EHA105(pSoup) is suitable for transgenic operation of rice, tobacco and other plants. The conversion efficiency was as high as 103CFU/μg by plant expression plasmid pGs2, and the conversion efficiency did not change after 12 months of storage at -70℃. |
Note | 1. When mixing the plasmid,it should be done gently,and the volume of the plasmid added should not be greater than 1/10 of the volume of the receptive state. The conversion efficiency will be sharply reduced if the plasmid is not pure or very large. 2. When there are too many colonies on the plate,the colonies are small. In order to obtain a large colony,the amount of plasmid used should be reduced or the amount of coating should be reduced,or the colony should be transferred to a new plate for growth. 3. The working concentration of rifampicin should not be higher than 25 μg/ml,too high rifampicin concentration will reduce the growth rate and conversion efficiency. 4. Rifampin has the effect of preventing the growth of miscellaneous bacteria and screening Agrobacterium;Adding streptomycin or gentamicin according to the resistance of the strain used can prevent the loss of Ti plasmid,but streptomycin is not conducive to the transgenic operation of Agrobacterium,so the addition of streptomycin or gentamicin can not be considered in the general culture of agrobacterium,and the probability of Ti plasmid loss is very low (negligible). |
