Protease S specifically cuts the peptide bond at the end of the -COOH group of aspartate or glutamate.

Precautions
Autolysis occurs at temperatures above 40℃. The enzyme was fully active in USP 0.2% SDS. In the presence of an ammonia buffer, the specificity of the enzyme may be limited to glutamyl bonds.
Unit definition: Using casein as substrate, at 37 ℃ and pH 7.8, one unit changes A280nm by 0.001 per minute.

Optimal pH: 4.0 and 7.8 (with hemoglobin as substrate)
Inhibitors: diisopropyl fluorophosphate (DFP) and univalent anions such as F-, Cl-, Br-, CH3COO- and NO3-
