Determination of Significance:
Methyl citrate synthase (MCS) is widely found in the mitochondrial matrix of animals, plants, microorganisms and cultured cells, and is involved in the regulation of the tricarboxylic acid cycle together with citrate synthase (CS).
Measurement Principle:
MCS catalyzes the production of methyl citric CoA by propionyl CoA and oxaloacetate, which is further hydrolyzed to produce methyl citric acid; This reaction results in the conversion of colorless DTNB to yellow TNB with a characteristic absorption peak at 412 nm, from which MCS activity can be calculated.
