In general, fluorescein substrates are more sensitive than coumarin or nitrophenol substrates. The fluorescein substrate is used to monitor cellulase activity. Cellulase is a class of enzymes containing beta-glucosidase, endoglucanase and extranase glucanase. These enzymes cut the beta-1, 4-D-glucoside bonds that connect glucose units containing cellulose. In addition to being produced by plants, cellulase activity is also found in many fungi and bacteria, including some plant pathogens. Most animal cells are not known to produce cellulase, in which cellulolytic activity is usually carried out by symbionts. The use of this sensitive fluorescein celloside makes it easy to monitor the activity of most cellulases. After cutting, the fluorescent compound fluorescein is released and activity measurements are easily obtained in a microtitration-plate based assay form.
