6-CDCFDA is a live cell fluorescent tracer probe, which can enter cells through passive diffusion and covalently bind to intracellular proteins, and is a long-acting cell tracer dye. Once inside the cell, the non-fluorescent 6-CDCFDA is hydrolyzed by lactase to produce green fluorescence. These fluorescent products can only accumulate in cells with an intact cell membrane, so dead cells without an intact cell membrane cannot be stained. 6-CDCFDA is not sensitive to pH changes and can be fixed by formaldehyde or glutaraldehyde.
Fluorescein diacetate (FDA) and its derivatives are non-fluorescent molecules that diffuse into cells and are hydrolyzed by intracellular non-specific esterases to produce fluorescent products. Fluorescent products can only accumulate in cells with intact cell membranes. As a result, dead cells are not stained. The dynamics of membrane transport and intracellular hydrolysis of FDA and its analogues, such as CDCFDA, are related to cell function, so FDA labeling can be used to monitor cells by flow cytometry or fluorescence microscopy. The fluorescence intensity of FDA-dye-labeled cells varied widely between cell lines and strains, possibly due to differences in cellular lactase activity.
