Mitochondrial red fluorescent probe is a red fluorescent dye containing chloromethyl functional groups that can label mitochondria. Ex/Em: 579/599 nm. Mitochondrial red fluorescent probes can be passively transported across the cell membrane and directly accumulate on the active mitochondria after simple incubation with living cells. The accumulation of the dye depends on the level of membrane potential. Once the mitochondria have been stained, they can also be fixed with an aldehyde fixative as required by subsequent experiments. For immunohistochemical and in situ hybridization experiments, cells need to be permeated first, and mitochondrial red fluorescent probes can also stain the mitochondria of permeated cells. The dye is suitable for double labeling experiments, and its red fluorescence can be well distinguished from other green fluorescence probes. Although traditional mitochondrial fluorescence probes such as TMR and Rhodamine 123 can easily be concentrated on functional mitochondria, they are washed off once the mitochondrial membrane potential is lost, thus limiting their application in some experiments that require cells to be immobilized with aldehydes or include factors affecting mitochondrial energy status.
Matters needing attention
1. Please centrifuge the product to the bottom of the tube instantaneously before use, and then conduct subsequent experiments.
2. For different cells and tissues, appropriate incubation time and staining solution concentration should be selected.
3. Fluorescent dyes have quenching problems, please try to avoid light to slow down fluorescence quenching.
