Trypan Blue is an anionic dye. Trypan blue can not penetrate the normal cell membrane of living cells, so living cells are not colored; The increased permeability of the cell membrane of dead cells can make dyes enter the cell through the cell membrane and make the dead cells colored blue, which is the most commonly used method to detect cell viability. The method could not distinguish between necrotic cells and apoptotic cells. When the cell is inactive or the cell membrane is incomplete, Trypan blue can stain it blue. Normal living cells have a complete cell membrane structure and can reject Trypan blue, and the cell will not be dyed blue. However, macrophages can phagocytose trypan blue, so it can be used as a living stain for macrophages.
Instructions (for reference only) :
1. Cell collection: Digest the adherent cells with pancreatic enzymes and/or EDTA, and the suspended cells can be collected directly. When the cells were collected, they were centrifuged at 1000-2000 rpm for 1 min, the supernatant was discarded, and the single-cell suspension was prepared and diluted appropriately.
2. Trypan Blue staining: Cell suspension is mixed with 0.4% Trypan blue solution at 9:1 ratio (so that the final concentration of Trypan blue is 0.04%), and the staining time is 3 minutes (generally, the dyeing time of 3 minutes is enough, and the dyeing time can be longer, but it should not exceed 10 minutes).
3. Cell count: Take an appropriate amount of stained cells and count them with a blood cell count plate. (Dead cells are blue and swollen, dull; Living cells do not stain and remain in their normal form, shiny.) Cell survival rate (%) = Total number of living cells/(total number of living cells + total number of dead cells) ×100%
Note:
1. The dyeing time should not be too long, otherwise living cells will gradually accumulate dye and dye color, so that the detection result is low.
2. If there is precipitation in the dye solution before dyeing, filter and remove the precipitation before use.
3. Trypan Blue is a potential carcinogenic hazard. For your safety and health, please wear a lab coat and disposable gloves.
