During cell apoptosis, a number of endonuclides are activated, which cut the genomic DNA between nucleosomes, producing 180 bp-200 bp DNA fragments, which are represented by the specific Ladder pattern shown in agarose gel electrophoresis. A lot of stickiness occurs when genomic DNA is double-stranded or single-stranded broken. The -OH terminal can be catalyzed by deoxyribonucleotide terminal transferase (TdT) and bind to SF/Cy-dUTP, so as to directly detect apoptotic cells by fluorescence microscopy or flow cytometry. Such methods are called Terminal-deoxynucleotidyl transferase mediated nick end labeling (TUNEL). Since normal or proliferating cells have few breaks in their DNA, there are no. -OH is formed and is rarely able to be stained. Tunel method can stain a single apoptotic nucleus or apoptotic body in situ, accurately reflect the typical biochemical and morphological characteristics of apoptosis, and can detect a very small number of apoptotic cells, so it is widely used in the study of apoptosis. This kit has a wide range of applications and can be used to detect apoptosis in frozen or paraffin sections, as well as in cultured adherent cells or suspended cells. It can selectively detect apoptotic cells, but not dead cells or cells with broken DNA strands caused by radiation and drug treatment. This kit can detect apoptosis in a short time, only one step dyeing reaction, and can be detected after washing.
Precautions
1. Please centrifuge the product to the bottom of the tube immediately before use, and then conduct subsequent experiments.
2. When the dyeing background is heavy or the non-specific coloring is obvious, the dyeing time can be appropriately reduced.
3. It is recommended to add negative control and positive control during the experiment.
4. Wear A mask and gloves when using component A. In case of contact with skin, rinse immediately with plenty of water.
5. Fluorescent dyes have quenching problems, please pay attention to avoid light as much as possible to slow down fluorescence quenching.
