Inhibitors of DNA viruses. Application in immunohistochemistry: BrdU labeled cell immunohistochemical staining (SP method), BrdU(bromodeoxyuridine) can be incorporated into the DNA strand synthesized by cells in S phase in vivo and in vitro to label DNA.
Application of Brdu in immunohistochemistry: BrdU labeled cells immunohistochemical staining (SP method), BrdU(bromodeoxyuridine) can be incorporated into the DNA strand synthesized by cells in S phase in vivo and in vitro to label DNA. The DNA synthesis cell frequency can be evaluated at the individual cell level by measuring BrdU incorporation by flow cytometry. "It is used to detect functional activation at the single-cell level, that is, DNA synthesis, expression of cell-surface activating antigens, and intracellular cytokine secretion." Activated mononuclear cells were included in the culture in the presence of BrdU, fixed and permeabilized, and BrdU uptake was measured with an anti-brdu fluorescent antibody. At the same time, the intracellular DNA was denatured with appropriate concentration of dnase to enhance the affinity of BrdU incorporation with antibody, while retaining the intracellular protein structure and fluorescence intensity of fluorescein. This method avoids the conflict between the conditions of DNA denaturation and the simultaneous detection of intracellular and surface markers. Continuous BrdU incorporation can be used to identify and analyze cells in the active state of DNA synthesis from those in the quiescent state, and thus to analyze the proportion of cell proliferation; On the other hand, the addition of BrdU at different time points allowed the analysis of cell cycle kinetics.
