Strain recovery
1. Solid and liquid media and culture equipment suitable for the growth of the strain should be prepared before resuscitating the strain, and all operations should be carried out under biosafety protection and aseptic conditions.
2. Take out the frozen storage tube and shake it vigorously to disperse the porcelain beads in the tube. Unscrew the frozen storage tube under sterile conditions, remove a small porcelain bead with sterile inoculation ring (inoculation needle) or tweezers, and tighten the cap.
3. Put the frozen storage tube back to the original storage environment as soon as possible. Excessive temperature change will weaken the viability of the strain.
4. The small porcelain beads taken out can be directly placed on the plate medium for marking inoculation, or directly placed in the suitable liquid medium for culture.
Strain preservation
The frozen storage tube after inoculation can be stored at -20℃ for 1 year and at -80℃ for 2 years.
