Product Introduction
Lambda exonuclease acts on double-stranded DNA, gradually cutting off 5′ single nucleotides in the 5′→3′ direction. The optimal substrate is 5′ phosphorylated double-stranded DNA, which also slowly degrades single-stranded DNA and non-phosphorylated substrates. The enzyme cannot start digestion from cuts or gaps in DNA.
Precautions
(1) 1×LambdaExoBuffer: 67 mmglycine-KOH (pH9.4 25℃), 2.5mMMgCl2, 50μg/mlBSA, incubated at 37℃.
(2) The optimum reaction temperature of this enzyme is 37℃, and it can be inactivated at 75℃for 10min.
(3) The cutting speed of the 5′-0H end is 20 times slower than that of the 5′-PO4 end, and single-stranded DNA is 100 times slower than double-stranded DNA.
