Dimethyl sulfoxide, English name DMSO. Often used as a dissolving reagent. Dimethyl sulfoxide (DMSO) is a sulfur-containing organic compound with a molecular formula of (CH3)2SO. It is a colorless, odorless, transparent liquid at room temperature and a combustible liquid with hygroscopicity. It has the characteristics of high polarity, high boiling point, good thermal stability, non-proton, miscible with water, and can dissolve in most organic compounds such as ethanol, propanol, benzene and chloroform. It is known as "universal solvent". Dimethyl sulfoxide (DMSO) cannot use containers made of PS, PVC and PC, but should use containers made of LDPE, HDPE and PP.
DMSO is a non-proton polar solvent, which is commonly used in chemical reactions, PCR reactions and vitrification cryopreservation in cells, tissues and organs. DMSO is used in cell freezing culture medium to protect cells from mechanical damage caused by ice crystals. It can also be used for cryopreservation of a series of cell lines, such as primary culture, subculture, recombinant aneuploid, hybridoma, embryonic stem cells (ESC) and hematopoietic stem cells. In addition, it is often mixed with BSA or FBS.
Uses:
(1) DMSO is widely used in low temperature protection of human and animal cell lines and bacteriophage λ;
(2) During cell fusion, 10% DMSO was added to 40-50% PEG solution;
(3) It has been reported that DMSO as a buffer component was used in the following experiments: purification of Poly (A +) RNA, transformation of E.coli competent cells, PCR reaction, amplification of cDNA library, DNA sequencing, DEAE-dextran mediated cell transfection, polycondensed amine mediated DNA transfection;
(4) There is a corresponding program that describes the recovery of DNA filtered by DMSO for subsequent PCR amplification;
(5) Capillary electrophoresis for DNA sequencing mixes 5% DMSO into 2M urea and adjusts the DMSO concentration to 100% if necessary;
(6) Some studies have reported the relationship between different concentrations of DMSO and the melting point temperature of oligonucleotides;
(7) Some studies have described the use of DMSO to increase the production of monoclonal antibodies in hybridoma cells; it was found that hybridoma cells were cultured at high temperature in a medium containing DMSO before cryopreservation;
(8) It has been reported that phosphoserine and threonine residues in DMSO modified protein were used as mass spectrometric analysis of phosphorylation state (MS);
(9) Literature reported the degradation of Leuprolide in DMSO and water.
