| In Vitro | Ganciclovir Sodium inhibits feline herpesvirus type 1(FHV-1)The median IC50 for feline herpesvirus type 1(FHV-1)inhibition was 5.2 micromolar，and no significant changes in morphologic characteristics，fusion，or survival of CRFK cells were observed at concentrations of the drug that were at or exceeded two times the IC50.[1] Ganciclovir concentration required to inhibit viral replication by 50 percent is 2.15 mumol.[3] The primary mechanism of ganciclovir action against CMV is inhibition of the replication of viral DNA by ganciclovir-5'-triphosphate(ganciclovir-TP). This inhibition includes a selective and potent inhibition of the viral DNA polymerase. Activity and selectivity are also amplified by the accumulation of ganciclovir-TP in CMV-infected cells.[4] |
| In Vivo | In adult rats，the intracochlear diffusion of ganciclovir(50mg/kg; 5days; i.p.)was shown to achieve the same concentration as in blood. Counts of white blood cells，red blood cells and platelets decreased significantly in ganciclovir-treated newborn mice.[2] Ganciclovir(1-80mg/kg/d; i.v.; 5days)dependently delayed MCMV-induced wasting syndrome and mortality，whereas a dose of 160 mg/kg per day induced reversible side-effects.[5] |
