Lipophilic fluorescent dyes PKH67 (Green) and PKH26 (Red) are suitable for conventional cell membrane labeling. The PKH26 fluorescent cell connection kit uses membrane labeling technique to bind yellow-orange fluorescent dyes with a long lipid tail to the lipid region of the cell membrane. The staining method depends on the type of cell and membrane, and is mainly used for cell labeling in vitro, cell proliferation in vitro and cell tracer in vivo and in vitro. The reagent kit provides the solvent (diluent C) needed in the dyeing process, which can increase the solubility and dyeing efficiency of dyes while maintaining cell vitality. Diluent C is isotonic to mammalian cells and does not contain detergents or organic solvents, nor does it contain normal saline and buffer salts. According to the cell type and the internal changes of the cell membrane after labeling, the surface of the labeled cells will change from uniform transparent to slightly protruding or patch-like. However, in the physiological range, the fluorescence of PKH26 was not affected by pH, and the fluorescence intensity of each cell had nothing to do with the location of dye labeling. PKH26 fluorescence is in the yellow-orange region (λex=551 nm, λem=567 nm), which can be used to label and track a variety of cells in vivo and in vitro. In cytotoxicity analysis, purple, green, red and far-infrared dyes emitted by fluorescent proteins, antibodies or DNA dyes in this region did not interfere with PKH26. PKH26 is most commonly used in dye dilution applications based on dye dilution proliferation analysis, including the establishment of a spectrum of antigen-specific precursors and the identification of resting or slow stem cells or progenitor cells in normal or tumor tissues. At the same time, PKH26 can also be used to monitor the uptake of exotic viruses, platelets and other nanoparticles; membrane distribution during stem cell division; cell-cell membrane transfer; cell phagocytosis; antigen presentation; adhesion; signal transmission through gap junctions; and neuronal migration in tissue sections. The fluorescence stability of PKH26 is very strong, especially when the research cycle of labeled cells is more than one week, PKH26 is used to track cells in vivo.
