Product Introduction
In the process of constant temperature amplification of nucleic acid molecules, whether LAMP, CPA and other amplification techniques are used, such high speed amplification characteristics will inevitably cause false positive amplification. The amplification of false positives is particularly obvious in LAMP amplification. Due to the high concentration of primers used in LAMP amplification, a small amount of mismatches and primer Dimer are inevitably guaranteed, and these slight contamination will lead to false positives in LAMP high-speed amplification. This product is optimized reaction buffer, combined with unique color development technology and negative control technology to establish the premix constant temperature mix series of products, which can be used for LAMP red and yellow color change, LAMP purple and green color change, LAMP orange and green (OG) color change with high sensitivity and high specificity. In LAMP amplification experiments, the Bst 4.0 MasterMix has faster amplification speed and impurity tolerance, thanks to the excellent performance of Bst 4.0 DNA/RNA polymerase, compared with the DNA/RNA LAMP MasterMix or LAMP Mix provided by other companies.
Bst 4.0 DNA/RNA polymerase has RNA template-dependent polymerase activity (reverse transcription) and DNA-dependent polymerase activity, so it can be used for efficient thermostatic amplification of both DNA and RNA samples. This product is an excellent reagent for LAMP and RT-LAMP amplification reactions. The optimized reaction system ensures the establishment of the reaction system of the detection reagent quickly.
This reagent is specially designed for OG orange green visual color-changing reaction. However, it can also be used to match other fluorescent dyes, such as SYBR Green I, Eva Green, SYTO-9, etc. Note that this reagent does not support HNB color development, turbidity, pH color change. Other detection methods are not tested. In addition, the reagent does not contain glycerol and can be used to establish a freeze-drying system.
Precautions
(1) OG dye tubes support a total reaction volume of 20-25 μl, and larger volumes may lead to a decrease in color concentration.
(2) As for the use of mineral oil, after preparing the LAMP reaction system, a drop of mineral oil can be added to cover the upper part of the reaction liquid to reduce aerosol pollution. Mineral oil does not affect the dissolution of OG dyes.
(3) In the use of other fluorescent dyes, the use of concentration can be optimized by itself, generally speaking, refer to the supplier to provide a concentration of 0.25-0.8x, too high concentration of dyes may lead to amplification failure.
(4) This reagent does not support turbidity, pH discoloration, HNB discoloration reaction, do not try.
(5) The reagent does not contain glycerin components, and the development of the lyophilized reagent may be carried out by persons with experience in lyophilization.
