Ponceau S, also known as scarlet, can be used for the detection of proteins on PVDF membranes, nitrocellulose membranes and cellulose acetate membranes. Ponceau is negatively charged and can bind to positively charged amino acid residues, while ponceau can also bind to non-polar regions of proteins to form red bands. The sensitivity of ponceau dyeing is less than that of Coomassie brilliant blue dyeing. The staining of proteins by ponceau is reversible and can be washed away with distilled water, PBS, or other appropriate solutions. Therefore, during protein N-terminal sequencing, the protein on SDS-PAGE glue was transferred to PVDF membrane, and the target strip was cut off for sequencing after staining with ponceumin.
Note: Ponceau staining solution is not suitable for the detection of proteins on nylon membranes. Ponceau dyeing solution can also be directly used to dye the PAGE glue, but it is best to soak the PAGE glue with methanol-acetic acid solution before dyeing to play a role in fixing the protein.
