AEC底物显色试剂是一种可以通过过氧化物酶催化底物3-氨基-9-乙基咔唑氧化形成红色不溶性产物,以此鉴定过氧化物酶活性,从而确定目标分子表达情况,该种方法权威而经典。其适用于辣根过氧化酶标记蛋白的免疫学检测(蛋白质免疫印记、免疫组织化学、以及原位杂交染色等)。产品性能稳定,操作便捷,敏感度高,显色清晰,重复性好。
3-氨基-9-乙基咔唑(3-amino-9-ethylcarbazole;AEC)是过氧化物酶(Peroxidase)的生色底物,其分子式为C14H14N2,分子量为210.3。在过氧化物酶的催化下,电子供体AEC底物在过氧化氢氧化下,失去电子而呈现出颜色变化和沉积,形成稳定性红色不溶性产物(能溶于有机试剂)。
说明:过氧化物酶活力指示剂;过氧化物酶底物,用于免疫组织化学染色;水溶荧光基因底物,用于纤维蛋白溶酶。适用于免疫印迹法和免疫组织化学中的染色步骤。该底物能生成不溶性红色终产物,可直观地观察到。用于生化试剂,荧光探针,标记,粒子和染色剂, 酶、抑制剂和底物等方面。
纯度 (滴定):>95.0%
溶解性:可溶于乙醇和DMF,参考浓度5%。
注意事项:
1. 由于AEC生成的颜色产物为脂溶性,因此用于IHC显色后不适合用中性树脂封片,须用水性封片剂封片也正因为如此,可以在显色后进行多重染色技术。
2. 根据情况自行优化实验条件,调整显色时间,获得最佳显色效果。
3. AEC有一定毒性,操作时请采取必要的防护措施。
