The amount of protease K required to hydrolyze substrate casein to 1 μmol tyrosine per minute at 37oC and pH7.5 is defined as one unit (U) of protease K enzyme activity. Effective in a wide pH range, the effective pH range is pH4.0-12.5, and the optimal pH range is pH7.5-8.0. The optimal reaction temperature of protease K is 65 ° C, but protease K itself is also very rapidly degraded at 65 ° C or higher. In many cases, the reaction temperature is chosen to be 50-55℃.

Protease K showed higher enzyme activity in the presence of 0.2-1%SDS or about 10mM urea. The common working concentration of protease K is 0.05-1mg/mL, and the specific working concentration is determined according to whether the buffer used contains SDS, urea, whether the pH is suitable, whether the temperature is suitable and other factors. Common concentrations of EDTA, Triton X-100, Tween 20, Sarkosyl and guanidine hydrochloride had little effect on the activity of protease K.
